Mechanism of action
A bactericidal aminoglycoside antibiotic. It binds to the 30S ribosomal subunit, causing misreading of the genetic code and inhibition of protein synthesis. It is effective against aerobic Gram-negative bacteria.
Indications
- Serious infections caused by susceptible aerobic Gram-negative bacteria
Contraindications & warnings
- Nephrotoxicity and ototoxicity (auditory and vestibular) — caution in renal impairment and dehydration
- May cause neuromuscular blockade
- Long tissue withdrawal periods — strongly considered in food animals
Drug interactions
- Nephrotoxicity is aggravated with loop diuretics, amphotericin, and some cephalosporins
- Enhances neuromuscular blockade with anesthetics and muscle relaxants
Prudent use of antibiotics
Prescribed based on diagnosis and susceptibility testing where possible, to limit antibiotic resistance.
